Purpose of the Test

A semen bacterial culture and sensitivity test with gram stain is performed to identify the presence of bacteria in the semen and determine the type of bacteria present. The test is used to diagnose and monitor bacterial infections of the male reproductive system, including prostatitis, epididymitis, and urethritis.

When this test is required

This test may be requested when a patient presents with symptoms of a bacterial infection of the male reproductive system, or as part of a routine fertility workup.

What the Test Detects

The test involves a microscopic examination of the semen sample to detect the presence of bacteria, followed by a culture and sensitivity test to identify the type of bacteria present and determine which antibiotics are most effective at treating the infection. A gram stain is also performed to identify the type of bacteria present based on their shape and staining properties.

Preparation for the Test

Patients may be advised to avoid ejaculation for 2-5 days prior to collecting the semen sample to ensure an adequate sample is obtained. Patients should also avoid using lubricants or condoms with spermicidal agents, as these can interfere with the test results.

Sample Requirements

The test requires a semen sample, which is typically collected by the patient through masturbation into a sterile cup.

Additional Notes

Patients should follow the healthcare provider’s instructions for collecting and handling the semen sample to ensure accurate test results. In addition, patients should inform their healthcare provider of any medications they are taking, as some antibiotics and other medications can interfere with the test results.
